De Novo Protein Design
De novo approach, also known as the inverse protein folding problem, involves designing proteins from scratch. De novo design has proven a powerful method for understanding protein folding and function, and for mimicking or even bettering the properties of natural proteins. De novo protein design mainly involves the generation of target backbone, the design of sequences whose lowest energy states are the target backbones, and the optimization followed by computational and experimental validation of selected sequence-structure pair.

CRISPR-Based Platforms
CRISPR (clustered regularly interspaced short palindromic repeat) is a segment of nucleic acid containing short, repetitive base sequences derived from DNA fragments of bacteriophages that had previously infected prokaryotic cells. These sequences, found in prokaryotic genomes, play an important role in prokaryotic defense. The prokaryotic immune system consisting of CRISPR and Cas (CRISPR-associated) proteins now form the basis of genome editing technology.

Cell-Free Protein Expression
Cell-free protein expression or cell-free protein synthesis (CFPS) refers to the protein expression approach that produces target proteins without using living cells. All the cellular machinery, such as ribosomes, enzymes, tRNAs, amino acids, and cofactors, are contained in CFPS for direct protein synthesis. A supplied nucleic acid template can be transcribed and translated in CFPS, and this process is not constrained by a cell wall or homeostasis conditions required to maintain cell viability.
